Main role
1. barrier effect
Grubauer and other reports have used acetone and other organic solvents to treat skin of male hairless mice at 1~3 months of age. The relationship between skin barrier function and skin lipids has been studied. The results show that skin barrier function decreases with the decrease of skin lipid content.
The relationship between Holleran, epidermal barrier function and sphingolipid synthesis into the [3H]H20 found that when the skin of hairless mice treated by acetone caused by skin barrier dysfunction, the sphingolipid synthesis increased in acetone after 5~7 h treatment, sphingolipid synthesis up to 170%; 24 h after the recovery of skin barrier function normally, sphingolipid synthesis also returned to normal. Thus, ceramide plays a very important role in maintaining the function of the skin barrier.
2. adhesion
Ceramide and cell surface proteins are linked by ester bonds to play the role of adhesive cells. This relationship is called "lipid, cell, and protein" package. Smith et al. Reported that the adhesion force between keratinocytes can be judged by determining ceramide content in epidermal cuticle, and the decrease of ceramide content in epidermal cuticle can make keratinocytes reduce adhesion and cause skin drying, desquamation and flake like.
Pawlmgs et al. Studied the mechanism of dry skin desquamation. It was found that the main reason for skin dryness is the decrease of ceramide content in the stratum corneum, and the content of ceramide is directly related to the degree of skin desiccation. The use of ceramide can obviously enhance the adhesion between keratinocytes, improve the degree of skin drying and reduce the skin desquamation. MaryKay Zhen Yan essence oil products contain such ingredients.
3. moisturizing effect
The water content of the skin is about 18% to 20% of the body weight. Skin moisture and elasticity are closely related to the moisture content of the epidermis. Excessive loss of skin moisture can lead to dry or chapped skin. Imokawa and other organic solvents were used to remove the cuticle lipid. It was found that the skin continued (>4 days) chapped, the scales were flaky, and the skin conductivity decreased significantly.
The skin components extracted from epidermis cuticle were subjected to local skin test under the same conditions. It was found that the local electrical conductivity of ceramide significantly increased the electrical conductivity of skin, followed by cholesterol and free fatty acids and cholesterol esters.
Ceramide has a strong association of water molecules, it through the structure to maintain skin moisture formation in the stratum corneum. Therefore, ceramide has the effect of keeping skin water.
4. anti aging effect
During the aging process of the skin, the lipid synthesis decreased and the content of ceramide in the cuticle decreased. The characteristics of skin aging are as follows: (1) dry skin, desquamation, rough and glossy: (2) thinning of the cuticle, increasing wrinkles and decreasing elasticity of the skin. Using ceramide can increase the ceramide content of stratum corneum, can improve dry skin, scaling, rough situation; at the same time, ceramide can increase the epidermal cuticle thickness, improve the capacity of the skin, reduce wrinkles and enhance skin elasticity, delay skin aging.
5. participates in cell function
Ceramide exists in animal and plant tissue cell membrane, the molecular monosaccharides or oligosaccharides residues exposed to the extracellular space in lipid head groups react with other cells, antibodies, bacterial toxins, viral envelope protein, thus ceramide in cell adhesion, cell immunity, blood type and decision as tumor antigen plays very important role. Neurosheathing (GSLs) is directly involved in cell recognition through the interaction of GSL - GSL and the interaction of GSL- protein agglutinin.
Nerve sheath glycosides, such as ganglioside GM, regulate the function of integrin receptors, and some special gangliosides directly regulate receptors, tyrosine kinases and protein kinase C (PKC) activities in the role of transmembrane signaling. Some neuroglycosides greatly promote the activity of receptor kinase, which leads to the enhanced activity of MAP kinase. Ceramide acts as a membrane antigen of some tumors, and is also a receptor for viruses, bacteria and toxins. They are the regulatory factors of the interaction of cell one cell, cell one microorganism, and cell adhesion protein.
6. regulation of cell growth and variation
The treatment of HL - 60 leukemia cells with C - one ceramide or other water-soluble derivatives can induce these cells to change. Exogenous ceramide has the same effect on HL-60 cells as ZNFot, 1, 25, one or two hydroxyl VD3 and interferon. C a ceramide (1 m) and its homologues have special anti HL one 60 cell proliferation effect, and have anti other leukemia cells and tissue culture of malignant cell activity; ceramide can regulate the growth and variation of nerve cells. 7. induced apoptosis
Ceramide is a cell toxin regulator in cells. In bone marrow and lymphocytes, ceramide derivatives cause the breakage of DNA in the early special nucleosomes, which are the external characteristics of cell apoptosis. Ceramide can regulate the apoptosis of TNFot cells and participate in other processes of apoptotic cells. The transcription factor AP 1 is the key to the apoptosis of HL 60 cells, and ceramide regulates the transcription of the signal by activating AP 1 1[4] and causes apoptosis.

Main application
1. in daily chemistry
Using ceramide containing Dynamisante essence cosmetics, can enhance the skin anti-aging function, keep the skin elastic, smooth and delicate, reduce facial wrinkles.
2. application in medicine
Ceramide plays an important role in inducing biological effects of various cytokines, vitamin D3, Fas and CD28 ligands. The role of El in cell apoptosis is also concerned. Ceramide is also involved in many physiological and pathological processes such as cell differentiation. In the process of epidermal keratinocyte culture, ceramide can induce apoptosis. Ceramide is the decomposed product of sphingomyelin in biofilm bilayer. It is recognized as the second messenger, and ceramide plays a wide and important role in cell growth, proliferation, differentiation, apoptosis and injury.
